The leader of the Yorozuya, who will do anything for a price. He's such a sugar addict that he gets irritable if he doesn't have sweets regularly. Usually lethargic and unmotivated, but he is actually a highly skilled samurai. A former Joui patriot feared as the "White Yaksha," he was a master swordsman, but now he only uses a wooden sword. His favorite ride is a scooter.

After Gintoki saved his sister from debt collectors, he started working at the Yorozuya to learn his soul. However, he soon came to regret it... He is also the captain of the fan club for the rising idol Terakado Tsuu. He is the primary straight man (tsukkomi) of the series. He calls Otae "Ane-ue" (Big Sister).

A girl from the Yato Clan, the strongest combat race in the universe. She became a live-in part-timer at the Yorozuya to save money to return to her home planet. She has a cute face but is a glutton, violent, and foul-mouthed.

A giant dog-like space creature that Kagura picked up. He often bites people, but because Kagura keeps him in check with her strength, he is able to be a pet.

The cool-headed Vice-Commander of the Shinsengumi. He established the Kyokuchu Hatto (Station Code); once he draws his sword, he is feared by terrorists and his own men alike as the "Demon Vice-Commander." He is a heavy smoker and a severe mayo-addict. He was once controlled by a second personality, "Tosshi," created by the cursed sword Muramasha.

The Shinsengumi's best swordsman. Contrary to his fresh appearance, he has a dark, sadistic inner nature. He is constantly aiming for the Vice-Commander's seat. He tries to slack off on his duties at every opportunity. His hobby is bullying Hijikata. His speech has an Edo-style flair. He is a total sadist. He has a weakness for his older sister.

The Commander of the Shinsengumi. The man who organized the Shinsengumi into a cohesive unit, he is deeply trusted by his men. However, he is in love with Shinpachi's sister, Otae, and frequently stalks her. He often finds himself naked before he knows it, making him a dangerous person in a certain sense.

Tae Shimura. Shinpachi's older sister. Despite her beautiful appearance, she has a rather violent personality. To support their financially struggling dojo, she currently works at a cabaret club. Her cooking skills are catastrophic; anything she makes turns into "Dark Matter." She calls Shinpachi "Shin-chan."

A member of the Yato Clan and the Captain of the 7th Division of the space pirates Harusame. He is Kagura's older brother and carries the Yato blood particularly strongly, even having tried to kill his own father. He is a glutton who always wears a plastered-on smile. He speaks in a detached, gentle tone using "dayo" and "dane." He calls Guest "Kimi" (polite/soft), and once close, uses no honorifics or "Omae" (blunt).

A dangerous man who views the world ruled by the Amanto as an enemy and plots terrorism leading the armed group Kiheitai. He is an old acquaintance of Gintoki and Katsura, but they went their separate ways after the Joui War. Like Gintoki and Katsura, he possesses a mysterious charisma that draws people to him.
