Born in late 19th-century England as the son of Dario Brando, Dio grew up in poverty under an alcoholic, violent father and a sickly mother. After his mother passed away early, young Dio resorted to any means necessary to earn a living. One day, he bought medicine for his father with money won from chess, but his father insulted him by throwing his mother's dress and demanding more alcohol. Enraged, Dio poisoned his father to death. Before dying, Dario left a letter entrusting his son to Lord Joestar. Dio became the Joestars' foster son and lived with Jonathan, but harbored ambitions to seize the family fortune and relentlessly tormented him. He harmed the family dog, cheated in boxing, and incited friends to isolate Jonathan, even going so far as to forcibly kiss Jonathan's love interest, Erina. Even after being defeated by an enraged Jonathan, he retaliated by burning Danny. Since then, he has hidden his true intentions, pretending to reconcile. Years later, now university students, the two are known as the pride of the school, but Dio is still targeting the inheritance and has been poisoning George Joestar's food. However, when Jonathan noticed the conspiracy through an old letter and confronted him, Dio became agitated, and Jonathan's suspicion turned into certainty. Since Jonathan lacks definitive evidence and needs an antidote, he has set out on a journey to Ogre Street, where Dio used to live. Dio briefly rejoices that he won't have to bother using the mask, but he is now wandering the streets, drinking heavily from a bottle, anxious that JoJo might return alive with the antidote. (Current progress) Later, Dio Brando fights with vagrants and puts the Stone Mask on one to test its power, only for the man to turn into a vampire and attack him. Barely surviving as the sun rises, Dio realizes the power of the Stone Mask (previously, he only planned to use it to kill Jonathan) and returns to the mansion, only to be surrounded by Jonathan and the police. Ultimately, he cries out, "I reject my humanity!" stabs Lord Joestar, and puts on the blood-stained Stone Mask to become a vampire himself.
